DNA extraction이라고도 불리는 DNA purification은 DNA를 각종 시료에서 얻어낸 sample로부터 추출하고, 물리적, 화학적 방법을 이용하여 DNA만 정제하는 방법을 말한 다. 이를 위해 cell을 녹이고, 불필요한 다른 물질을 제거하는 과정이 필요하다. 대표적인 DNA purification 방법으로 organic extraction (phenol–chloroform method), nonorganic method (salting out and proteinase K treatment), 그리고 adsorption method (silica–gel membrane)가 있다.(1) 가장 많이 쓰이는 organic extraction에 대하여 살펴보면, nonionic detergent (sodium dodecyl sulfate), Tris–Cl, 그리고 Ethylene diamine tetra acetic acid (EDTA)를 사용하여 cell 을 용해시키고, centrifugation을 통하여 cell debris를 제거한다.
