Hemocytometer의 원리를 이해하고, 이를 통해 실험에 필요로 하는 적정량의 세포 수를 계산할 수 있다.

5. Materials
MCF-7 Cells (cultured in T25 flask), DPBS, 0.05% Trypsin-EDTA solution, Media (RPMI1640, FBS, P/S), alcohol lamp, pipette aid, serological pipette, incubator, conical tube, centrifuge, micropipette, tip, rack, beaker, 1.5㎖ ep-tube, trypan blue solution, hemocytometer, 35㎜ dish, 70% ethanol, tissue, clean bench, 광학현미경, 계수기

6. Methods
① 4℃에서 냉장보관 중이던 media를 37℃ water bath에서 충분히 pre-warmed.
② Clean bench에 넣기 전, pre-warmed media 및 필요한 실험기구들을 70% 에탄올로 소독한다. (Cell이 들어있는 flask는 에탄올로 소독하지 않고 clean bench로 옮겨준다.)
③ 세포주를 배양 중인 flask 내부의 media를 pipette aid를 이용해 aspiration & discard.
④ DPBS washing: DPBS 3㎖을 flask에 조심히 분주하고 좌우로 gently washing 해준 후, DPBS를 다시 aspiration & discard.
